bengal, asparagus agar (Allen, 1951), asparagus agar plus 1:80,000 crystal violet, and asparagus agar plus 1:15,000 rose bengal. The addition of dyes decreased the number of contaminants but also appeared to inhibit the development of the rhizobia. All plates were examined daily for 2 weeks. Smears of colonies which resembled rhizobia were examined. Pure cultures were obtained by the use of both pour and streak plates. These pure cultures were inoculated into Bacto-MR-VP Medium', alkaline, and very alkaline broth (Hofer, 1935). At the same time, known pure cultures of rhizobia were inoculated into the same media. The reactions of the isolates are given in table 1. After 1 to 2 years' storage under mineral oil at room temperature or refrigerator temperature, all nodules were dark and soft. No typical rhizobia were isolated from nodules stored under mineral oil. The

Effect of inoculum preparation and dietary energy on microbial numbers and rumen protein degradation activity.

Chilling of whole rumen contents prior to preparation of strained rumen fluid enriched with particle-associated microorganisms resulted in rumen inoculum with the highest microbial dry weight, total bacterial counts, and degradation rates for casein and soybean meal. Strained rumen fluid, whole rumen contents blended with strained rumen fluid, chilled strained rumen fluid, and strained rumen fl...

Proteolytic activity of rumen microorganisms and effects of proteinase inhibitors.

Proteolytic activity of the bovine rumen microflora was studied with azocasein as the substrate. Approximately 25% of the proteolytic activity of rumen contents was recovered in the strained rumen fluid fraction, and the balance of the activity was associated with the particulate fraction. The proportion of proteinase activity associated with particulate material decreased when the quantity of ...

Next Generation Sequencing to Define Prokaryotic and Fungal Diversity in the Bovine Rumen

A combination of Sanger and 454 sequences of small subunit rRNA loci were used to interrogate microbial diversity in the bovine rumen of 12 cows consuming a forage diet. Observed bacterial species richness, based on the V1-V3 region of the 16S rRNA gene, was between 1,903 to 2,432 species-level operational taxonomic units (OTUs) when 5,520 reads were sampled per animal. Eighty percent of specie...

Investigating the Effects of Cadmium on Rumen Microbial Fermentation and Nutrient Digestibility Using Gas Production

Effects of different levels of cadmium (0.1, 1, 2, 4, 8, 16, 32, 64, 128, 256 mg/l of culture medium) were evaluated using gas production technique. In the glass vials used to measure gas production 30 mg of buffered rumen fluid (2:1 ratio of rumen fluid: buffer) poured in glass and cultured at 38.6 temperature. Volume and pressure of produced gas were measured with syringes and digital pressur...
